医薬品開発における動物試験代替法として、ヒト培養細胞を用いた評価(細胞アッセイ)が注目されている。生体外で細胞の臓器特異的機能を発現させることを目指して我々は、培地中で自立的に浮遊するハイドロゲル膜を細胞の培養足場として用いたスフェロイド培養系を構築した。基材に固定された培養と比較して本培養系では、細胞のグルコース消費量に対する乳酸産生量の減少が確認され、好気的な培養環境が構築されていることが示唆された。さらに、薬物代謝酵素CYP1A2に関する遺伝子発現量の向上が確認され、本培養系はHepG2の肝特異的機能の発現に寄与することが示唆された。
As one of the alternatives to animal experiments in the drug development, bioassay using cultured human cells (cell-based assay) has been gaining increased attention. Aiming to express organ-specific functions of cells in vitro , we constructed cell spheroid culture system using flexible hydrogel membrane floating in the culture medium as cell culture scaffold. Compared to the cell culture system immobilized on the substrate, lactate production against glucose consumption decreased, and it was suggested that aerobic culture environment was constructed on the floating membrane. Moreover, gene expression level of CYP1A2, which is one of the drug-metabolizing enzymes, was enhanced, and this result suggests that the spheroid culture system on the floating hydrogel membrane expressed liver-specific functions of HepG2 cells.

一部が基材に固定されたまま培養液中で自立的に浮遊するやわらかなハイドロゲル膜を足場とする細胞集塊(スフェロイド)培養系を構築した。本培養系でHepG2を培養したところ、従来の基材に固定された単層培養と比較して、肝特異的な薬物代謝酵素に関する遺伝子発現量の向上が確認された。
We constructed cell spheroid culture system using flexible hydrogel membrane floating in the culture medium as cell culture scaffold. When HepG2 cells were incubated in this culture system, it was shown that gene expression of the liver specific drug metabolizing enzyme was enhanced compared to conventional monolayer cell culture system on the substrate.

従来のディッシュ培養では大幅に抑制される細胞機能の発現を目指して、一部が基材に固定されたまま培地中で自立的に浮遊するやわらかな細胞接着性ハイドロゲル膜を創製し、このゲル膜を培養足場とする新規細胞培養系を開発した。
Aiming at expression of physiological cell functions which are suppressed drastically in the conventional dish cell culture, we developed a novel cell culture system using flexible cell-adhesive hydrogel membrane floating in the culture medium as cell culture scaffold.
